The melanocortin peptide Melanotan II

Melanotan II is a cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), engineered for superpotency and resistance to enzymatic degradation — a non-selective agonist of the melanocortin receptors MC1R through MC5R. It is a central tool compound in pigmentation and melanocortin-pathway research.

Research spans melanogenesis and photoprotection, appetite and energy-balance signalling through MC4R, and sexual-function pharmacology, the pathway from which its metabolite bremelanotide was later developed. RESEARCH BACKGROUND

The first superpotent melanotropic peptide

In the 1996 phase-I pilot study at the Arizona Cancer Center, Melanotan II induced visible skin darkening in fair-skinned volunteers after only five low subcutaneous doses, without ultraviolet exposure. In bioassays the cyclic lactam-bridged peptide proved far more potent and longer-acting than natural alpha-MSH.

Follow-up work characterised its effects on erectile function — leading to the development of its metabolite bremelanotide (PT-141) — and made it a reference agonist for mapping the melanocortin receptor family across pigmentation, appetite and sexual function.
